Don't want to discourage you but poison assassin just doesn't deal enough damage - played it in 3.15, and the damage hit a wall at T15+, the "spend 20+ EX or you're not progressing past this" type of wall, and it was so bad at bosses even 50 EX wouldn't fix it

I've reached the same conclusion, right now both CL and VG are in a really bad spot, either GGG makes changes to compensate the damage loss in order to let us build up defenses or vice-versa.

Both the poison nerf and the changes to defenses were just uncalled for...

K, this is how far I've got, this is with Venom Gyre but since I've adapted OP's build to it, I think this could be changed a bit to work with Cobra Lash, only done Conquerors so far, even when taking hits it's manageable, boss damage it's just frustrating, probably close to a minute or so to kill each conqueror, the investment is just ludicrous.

I'll probably mess a bit with the flasks since I haven't touched them.

I'll probably cry when I try Maven/Shaper.
