do minions use life flask ?

I play perfect druidic alchemists spectres, with Umbilicus immortalis belt. My spectres throw poisonous concoction (spell ? not attack?). Poisonous concoction description: "an attack skill gem, charges can be consumed from a life flask to add further damage".

Will my spectres use my life flask charges to increase damage? Should I actively use life flask myself, or they take charges automatically?

They will not use your flask charges for it.

You need to equip a life flask in your main hand and corrupt it with a "Chains +1 times" modifier, then start throwing life flasks at your spectres.
