All of these challenges require that the map instance (not the map item) have Increased Item Quantity.

In your screenshot, the map has 0% IIQ and 666% IIR, indicating that you have Meticulous Appraiser allocated.
You cannot progress these challenges while Meticulous Appraiser is allocated.
